چکیده

We have developed amicrorheometer, based on optical tweezers, in which hydrodynamic coupling between the probe and fluid boundaries is dramatically reduced relative to existing microrheometers. Rotational Brownianmotion of a birefringentmicrosphere within an angular optical trap is observed bymeasuring the polarisation shifts of transmitted light. Data gathered in this manner, in the strongly viscoelastic fluid Celluvisc, quantitatively agree with the results of conventional (bulk) rheometry. Our technique will significantly reduce the smallest sample volumes which may be reliably probed, further extending the study of rare, difficult to obtain or highly nonhomogeneous fluids.

Microrheology from rotational diffusion of colloidal particles.

The microrheology of viscoelastic fluids is obtained from rotational diffusion of optically anisotropic spherical colloidal probes, measured by depolarized dynamic light scattering. The storage and loss moduli obtained from the rotational mean squared displacement is in excellent agreement with those obtained from translational diffusion and by mechanical measurements. We also show that this me...

Pulsed laser manipulation of an optically trapped bead: averaging thermal noise and measuring the pulsed force amplitude.

An experimental strategy for post-eliminating thermal noise on position measurements of optically trapped particles is presented. Using a nanosecond pulsed laser, synchronized to the detection system, to exert a periodic driving force on an optically trapped 10 μm polystyrene bead, the laser pulse-bead interaction is repeated hundreds of times. Traces with the bead position following the prompt...

Green fluorescent protein in inertially injected aqueous nanodroplets.

We inertially inject and study the contents of optically trappable aqueous nanodroplets (hydrosomes) emulsified in a perfluorinated matrix. A new piezoelectric actuated device for production of single hydrosomes on demand is introduced. Hydrosomes containing enhanced green fluorescent protein (EGFP) were injected, optically trapped, and held at the focus of an excitation laser in a confocal mic...

Microrheology with Optical Tweezers: Measuring the relative viscosity of solutions ‘at a glance'

We present a straightforward method for measuring the relative viscosity of fluids via a simple graphical analysis of the normalised position autocorrelation function of an optically trapped bead, without the need of embarking on laborious calculations. The advantages of the proposed microrheology method are evident when it is adopted for measurements of materials whose availability is limited,...

Rotational diffusion of spherical colloids close to a wall.

There is currently no experimental technique available to probe spatially resolved rotational diffusion of nanoparticles in the vicinity of a wall. We present the first experimental study of rotational diffusion of small spherical colloids, using dynamic evanescent wave scattering. A setup is used where the wave vector components parallel and perpendicular to the wall can be varied independentl...
